Cancer genomes:
One of the research lines addresses the study of the molecular mechanisms involved in tumour transformation or the development of resistance to treatment. Through the sequencing and analysis of complete genomes of tumour samples, as well as the development of bioinformatics tools, the group has contributed to defining the main genomic alterations involved in the development of different types of leukaemia and lymphoma. It is currently applying single-cell sequencing techniques to understand the mechanisms of communication between tumour cells and cells in the tumour microenvironment. Together, these approaches have made it possible to identify dozens of cancer driver genes, which, together with the development of bioinformatics tools, allow progress to be made in genomic precision medicine in oncology. Work is also being done to understand the mechanisms by which these new cancer genes contribute to tumour transformation through the development of new preclinical models and their functional and molecular characterisation.


Molecular basis of ageing:
Ageing constitutes the main risk factor for the development of cancer, so knowledge of the molecular mechanisms involved in this process is of great interest in the context of oncology. Our group has been characterising the molecular mechanisms altered during ageing, as well as its associated pathologies, paying special attention to the relevant aspects in relation to cancer biology. Through the development of animal and cellular models of accelerated ageing, the main biochemical, metabolic and molecular pathways altered in this process have been determined, and therapies based on gene editing have been developed for the treatment of these alterations. We are also investigating the mechanisms involved in the presence of age-associated clonal haematopoiesis in adults, which leads to an increased risk of developing different types of leukaemia, and developing mathematical models to identify individuals at greater risk of transformation.
GROUP HEAD BIOGRAPHY
Xose S. Puente is Professor and Head of the Department of Biochemistry and Molecular Biology at the University of Oviedo. D. in 1998, he did a postdoc at The Scripps Research Institute (USA) on the role of proteases in cancer. Since 2003 he has been developing a line of research in comparative genomics and bioinformatics, participating in large international sequencing consortia. He has contributed to the development of tools for detecting tumour mutations, revolutionising our knowledge of chronic lymphocytic leukaemia by identifying more than 60 genes that are drivers of this pathology. Some of these mutations affect non-protein coding regions of great functional relevance, such as the NOTCH1 gene or the U1 snRNA. He has published more than 150 articles in journals such as Nature, Nature Genetics and Leukemia. He co-founded the company DREAMgenics and is actively involved in teaching, training young researchers and dissemination.
